Formulation The IgG fraction of culture supernatant was purified by Protein A/G affinity chromatography and lyophilized from a 0.2 µm filtered solution in phosphate-buffered saline (PBS, Azide Free).
Reconstitution and Storage Reconstitute the antibody with sterile PBS and the reconstituted antibody can be aliquoted and stored frozen at < -20 for at least for six months without detectable loss of activity. Avoid repeated freeze-thaw cycles. Lyophilized samples are stable for 2 years from date of receipt when stored at -70°C.

In Vitro CD40 Activation

(1) Purified mouse splenic B cells are stimulated with gAP-0012 for 24-48 hrs,
(2) Activation markers such as CD86, CD80, and MHC class II are significantly upregulated compared to untreated cells

Blocking of CD40-CD154 interaction

(1) Purified mouse splenic B cells express CD40
(2) Incubate B cells with variable doses of gAP-0012 together with a fixed concentration of recombinant mouse CD154-fused with HIS-Tag
(3) gAP-0012 dose dependently reduces the binding of CD154-HIS protein
